Source/origin paths and follower paths
I'm looking for a way to set a certain layer/path as the origin so when I duplicate and change the original path, the rest will automatically update.
This would for example be useful in iterative design workflow to quickly change designs and/or clipping mask instances where you need multiple copies of the same path, but only wish to edit one

Illustrator offers two ways to achieve this already.
One is Symbols, which converts a desired object into a source graphic, which you can later use anywhere in the document. Once you change the symbol, the copies of it get updated.
The other one is Global Edit (Select > Start Global Edit, or a button which says 'Start editing similar shapes together' in the Control panel or 'Start Global Edit' in the Properties panel).
Both have some limitations, but hey fit into your request enough to think of it as closed. Do these work for you?
