Opacity and Mask Define Knockout Shape - This option does not work if GPU Preview is enabled.
Please get in touch with us at any of the other support channels – https://helpx.adobe.com/support.html . Since this is not a generic issue that we can reproduce at our end, we will need someone to look into your machine to figure out what is going on here.

I have also encountered this issue.
Please find attached a file that demonstrates different behavior in GPU preview. The file has a Group(KO) containing two rectangles - green(50% opacity and Opacity & mask define KO shape enabled) and red. When GPU preview is enabled, enabling the Opacity & mask define KO shape option has no effect on the rendering and green knocks out red.
