Opacity and Mask Define Knockout Shape - This option does not work if GPU Preview is enabled.

I have also encountered this issue.
Please find attached a file that demonstrates different behavior in GPU preview. The file has a Group(KO) containing two rectangles - green(50% opacity and Opacity & mask define KO shape enabled) and red. When GPU preview is enabled, enabling the Opacity & mask define KO shape option has no effect on the rendering and green knocks out red. -
