Setting Opacity twice wrongly affects the CMYK values on effects underneath
(2019 Mac Pro - Sequoia 15.6.1 - Illustrator 29.8.2)
Draw a box, fill it with 25% black, apply an effect such as Blur through the Appearance Panel.
On a new layer, draw another box which intersects the previous box. Make this new box 50% Magenta. Set it to Darken.
Select the newly created layer and change the blending mode on the entire layer. Set it to Darken as well.
View Separations Preview. Black only. You will notice that the MAGENTA box has intensified the BLACK plate in the areas where it intersects.
This happens when choosing any CMYK color, not just magenta on black. It also happens if you double any blending mode - Darken, Multiply, etc. Even Overprinting and Darkening the same object affects the underlying effect.

Yes, does the same thing for me.
I remember some other reports about doubled blending mode getting calculated wrong. Will merge or link if I found any.
